Commonly abbreviated to Harrisburg Lancaster Railroad, it was one of the oldest American railway companies, obtaining a government concession to build its 52-mile line back in 1832. The railway was opened as early as 1838 and from 1861 the operation of the line was contracted out to the Pennsylvania Railroad Company, from which the company was later absorbed.

The company operated a 36-mile line from Harrisburg to Dillerville, with a station in Columbia. In 1860 the railway was leased to the Pennsylvania Railroad for 999 years. John Edgar Thomson was executive director and financier of the railway. He was the third president of the Pennsylvania Railroad. Thomas Scott was one of the great railroad personalities of his time. After his attempt to build a railway line through the southern United States failed due to Collis P. Huntington and his Southern Pacific, Scott took over the management of the Pennsylvania Railroad in 1874. During the 1970s, he expanded it into the largest railroad system close to that of the Vanderbilts (Penn-Central System). Two years earlier he had already become president of the large Pennsylvania Company, the holding company of the Penn-Central System.
